HDR templates that mediate genome repair can either be double or single-stranded DNA.
Using our myDNA™ technology we can synthesize either plasmid or long ssDNA HDR templates. Multiple reports have demonstrated that ssDNA is generally more efficient at mediating HDR than dsDNA templates. It can be difficult to make ssDNA of a length over 200 bases and many companies are unable to synthesize such molecules. Our unique DNA synthesis methods allow us to routinely make ssDNA of high sequence fidelity and purity at sizes greater than 200 bases in length. These long ssDNA templates allow researchers to place entire genes into their genomic cut site and greatly increases the sophistication of genome editing which can be accomplished at a single cut site. HDR template synthesis is possible regardless of model organism, so long as a break in the genome can be made an HDR template can be designed to mediate HDR. All HDR templates are subjected to strict QC procedures, including being fully sequenced by next-generation sequencing, so that we deliver high fidelity and highly uniform DNA molecules for each application.
