myTags – FISH Probes
myTags Expert SARS-CoV-2
FISH probe set for localization of full-length RNA sequence of the 2019 novel coronavirus (SARS-CoV-2) responsible for COVID-19 disease.

The myTags® technology has been expanded to development of FISH probe sets to detect full-length sequence of both the native viral RNA or positive (+) strand and the actively replicating RNA or negative (-) strand from the SARS-CoV-2 virus. The two probe sets were designed to detect both strands in the same hybridization reaction or separate reactions. These probe sets may be used in various FISH applications including human and Vero (African Green Monkey kidney epithelial) cell cultures.

The myTags Expert SARS-CoV-2 probes were designed using the most recent genome available.  These probe sets are specific to the SARS-CoV-2 virus and will not hybridize significantly to any other circulating human coronaviruses. The probe set is provided as either myTags Immortal, which can be amplified for continual use and labeled by the end-user, or as myTags Labeled probes which arrive ready for immediate use in cytogenetic applications.

Probe Coverage

The myTags Expert SARS-CoV-2 probes are available to detect both the positive (+) strand and negative (-) strand sequences. The (+) probe set will detect the viral genomic RNA. Detection of the virus (-) strand is indicative of active viral replication. The design strategy used to generate these probes enables the researchers to use the both sets together or in separate hybridization mixtures as probes in each set will not hybridize to one another.



Performance

Highly Specific Detection of the SARS-CoV-2 Viral Genome

Overview

myTags Expert SARS-CoV-2 probes are available to detect both the positive (+) strand and negative (-) strand sequences. SARS-CoV-2 is a class IV RNA virus with an ssRNA genome (positive strand). The probes to detect the (+) strand will hybridize to full-length genomic (+) strand RNA as well as subgenomic (+) strand RNAs made during replication. Detection of the (-) strand is a hallmark of active viral replication. The probes to detect the (-) strand will hybridize to the full-length (-) strand as well as subgenomic (-) strand RNAs.

The full-length SARS-CoV-2 genome sequence was used for probe design. These probes are designed to not significantly detect other circulating human coronaviruses, including beta, alpha, and delta coronaviruses. The design strategy used for these probes enables the researchers to use the both sets together, or in separate hybridization mixtures, as probes in each set will not hybridize to one another.

These probes can be used in various FISH applications. The probe sets have been designed to be used in human cell lines and tissue, and have been confirmed for application in Vero cell cultures. A suitable application of these probes would be cell culture experiments utilized in drug discovery research. In experimental applications where viral infection is not controlled, additional confirmation of SARS-CoV-2 presence via PCR is recommended.

myTags Expert SARS-CoV-2 probes are available two formats: Immortal probes that are ready to be coupled with your preferred tag(s), or ready-to-use Labeled probes with Alexa-488, ATTO-550, or ATTO-647N fluorescent tags. For experiments that need a brighter signal, the 3X fluorescent tag upgrade is recommended. The brighter signal from the triple tags can assist in detection of lower cellular viral RNA concentrations at earlier experimental time points.
