myTags – FISH Probes
myTags® Expert SARS-CoV-2
myTags® FISH probe set for localization of full-length RNA sequence of the 2019 novel coronavirus (SARS-CoV-2 / 2019-nCoV) responsible for COVID-19 disease.

The myTags® technology has been expanded to development of a FISH probe sets to detect full -length sequence of both the native viral RNA or positive (+) strand and the actively replicating RNA or negative (-) strand from the SARS-CoV-2 virus. The two probe sets were designed to detect both strands in the same hybridization reaction or separate reactions. These probe sets may be used in various FISH applications including human and Vero (African Green Monkey kidney epithelial) cell cultures.

The myTags Expert SARS-CoV-2 probes were designed using the most recent genome available.  These probe sets are specific to the SARS-CoV-2 virus and will not hybridize significantly to any other circulating human coronaviruses. The probe set is provided as either myTags immortal, which can be amplified for continual use and labeled by the end-user or as and myTags labeled probes which arrive ready for immediate use in cytogenetic applications.

Cell cultures experiments utilized in drug discovery are examples where myTags Expert SARS-CoV-2 probes can be used. Experiments that need a brighter signal can be visualized using the 3X fluorescent upgrade tags. The brighter signal from the 3X tags can detect lower viral RNA concentrations in the cells during earlier time points.




Applications

The full SARS-CoV-2 genome sequence was used to generate the myTags SARS-CoV-2 probes.  These probes will not significantly detect other circulating human corona viruses.   including a collection of beta, alpha and delta coronaviruses.  In applications where the virus infection is not controlled, confirmation of the presence of SARS-CoV-2 by PCR is recommended.

The SARS-CoV-2 is a class IV RNA virus.  The genome for this virus is a ssRNA (+) strand.  The myTags Expert SARS-CoV-2 probes to detect the (+) strand will hybridize to full-length genomic (+) strand RNA  as well as subgenomic (+) strand RNAs made during replication.  Detection of the (-) strand is a hallmark of virus replication.  The  myTags Expert SARS-CoV-2 probes to detect the (-) strand will hybridize to the full-length (-) strand as well as subgenomic (-) strand RNAs.
