Daicel Arbor Biosciences, a division of Chiral Technologies, Inc and worldwide leader in next generation sequencing (NGS) target enrichment, is joining the International Wheat Genome Sequencing Consortium (IWGSC) as a sponsoring partner.

The IWGSC is an international, collaborative consortium of wheat growers, plant scientists, and public and private breeders dedicated to the development of genomic resources for wheat scientists and breeders. These resources aim at facilitating the production of wheat varieties better adapted to environmental challenges, with higher yields, enhanced nutritional quality, and improved sustainability.

Sponsors are an essential part of the IWGSC. They participate in IWGSC-led projects and, as members of the Coordinating Committee, they help shape the IWGSC priorities, strategic plans, and activities. Jacob Enk, Ph.D., NGS R&D Manager, will represent Daicel Arbor Biosciences in the IWGSC Coordinating Committee.

Chinese Spring, IWGSC RefSeq v1.0, which was fully annotated and published by IWGSC members. The panel utilizes over two million probes to cover 200 megabases of high confidence exons in the genome and is compatible with both hexaploid and tetraploid cultivars of wheat. Daicel Arbor Biosciences is currently developing new iterations and expansions of the exome panel based on requirements of IWGSC members.

– Swift Biosciences and Daicel Arbor Biosciences, a division of Chiral Technologies, jointly announced a partnership to offer a complete workflow for RNA-Seq library preparation and targeted enrichment of the SARS-CoV-2 genome. An enabling offering for epidemiological research and public health surveillance, the partnership leverages Swift’s patented Adaptase® technology within the Swift RNA Library Kits combined with myBaits technology from Daicel Arbor Biosciences for hybridization-based target enrichment of the virus for whole genome sequencing.

Hybridization-based target enrichment can retrieve target sequences that have significant genomic rearrangements or mutations relative to the reference used for probe or primer design. This allows the Swift and Arbor workflow to be robust to structural nucleotide variants segregating within the SARS-CoV-2 population.

“We have been providing the SARS-CoV-2 panel free of charge to research labs studying the evolution of the novel coronavirus and are excited to partner with Swift Biosciences on this effort,” stated Alison Devault, Director of Genomics at Daicel Arbor Biosciences. “Detection of viral mutations is heavily reliant on sample preparation prior to sequencing and Swift’s RNA library prep meets the sensitivity required for this virus.”

The two companies’ combined product offering includes modules for RNA fragmentation and reverse transcription, library preparation and amplification, indexed adapters, capture probes, blockers, and wash buffers for SARS-CoV-2 enrichment. No ribosomal RNA depletion or other enrichment or depletion steps are required. Customers in North America may purchase the full set of reagents directly from either Swift or from Daicel Arbor Biosciences.

Overview of the Joint Partnership

  • An effort is underway for the development of a prophylactic DNA vaccine for the SARS-CoV-2 virus between Osaka University (Department of Clinical Gene Therapy/Department of Health Development and Medicine), and AnGes. Inc., utilizing AnGes, Inc.’s plasmid DNA platform.
  • The DNA vaccine production process is generally established in a shorter period of time than the production of conventional vaccines, and is being performed by Takara Bio Inc., who has the plasmid DNA production technology and facilities.
  • DAICEL has developed a gene transfer method using the ActranzaTM device, and is conducting research for its clinical application with Osaka University (Department of Device Application for Molecular Therapeutics/Department of Health Development and Medicine). This technology enables efficient delivery of plasmid DNA encoding antigen protein directly into cells via intradermal delivery, in order to initiate antibody production in the body.

About DNA Vaccines

DNA vaccines can be produced in a short period of time without pathogenic components. By inoculating a person with DNA plasmid encoding for a specific viral protein of the pathogen, the antigen protein is produced in the body without effects of the virus, while generating an immune response and thus further immunity against the pathogen.

Leveraging their expertise in designing and manufacturing high performance NGS enrichment panels for pathogens, Daicel Arbor Biosciences produced the new 2019-nCoV panel in just one week following a request from the research community. To aid in urgent research on this global threat, Arbor will provide complete 2019-nCoV hybridization capture kits free of charge to researchers worldwide who are studying the epidemiological and phylogenetic properties of this novel virus.

New myTXTL bulk packaging align with volumes required for high-throughput organizations in synthetic biology.

Daicel Arbor Biosciences, a division of Chiral Technologies, Inc. and leader in developing molecular biology research tools, today announced the launch of myTXTL® bulk pack sizes while attending the SynBioBeta 2019 conference in San Francisco, CA, as an addition to their growing catalog of cell-free expression kits for synthetic biology applications. This new offering aligns with the growing demand for larger volume requirements by organizations utilizing high-throughput screening in protein and enzyme engineering. The pack sizes and aliquots of the master mixes are ideal for cell-free expression on automated liquid handling systems.

Cell-free expression systems are currently available from Daicel Arbor Biosciences as a Sigma 70 or a linear DNA expression master mix. Both systems use E. coli as a base lysate combined with amino acids, essential elements, and a regenerative energy buffer in a complete, simple to use master mix which only requires the addition of template DNA. The Sigma 70 system relies on circular or plasmid DNA while the linear system can transcribe circularized or linear DNA as a template prior to translation. Either system will be available to researchers in bulk format to pair with their high-throughput DNA template generation systems.

Sequencing and analysis of the entire bread wheat genome is incredibly difficult and costly due to its enormous size, complex sub-genomes, and more than 85% of the genome being composed of repetitive elements. Daicel Arbor Biosciences, a worldwide leader in next generation sequencing (NGS) target enrichment and synthetic biology, announces their partnership with the International Wheat Genome Sequencing Consortium (IWGSC), an international organization dedicated to developing a gold-standard reference genome for bread wheat cv. Chinese Spring. This strategic partnership will provide IWGSC members with a standardized exome panel for research and development as well as expand Arbor’s collaboratively developed myBaitsÒ Expert panels in agrigenomics.

“Our members have been working together since 2005 to generate a gold-standard reference genome for Chinese Spring.” said Kellye Eversole, Executive Director of the IWGSC. “Partnering with Daicel Arbor Biosciences will accelerate research efforts into the inner-workings of this very complex genome.”

Sequencing the bread wheat genome was an incredible feat, due to its enormous size – five times larger than the human genome – and complexity – bread wheat has three sub-genomes and more than 85% of the genome is composed of repeated elements. The potential to analyze only the exome, or just the functionally expressed genes, greatly reduces the cost of sequencing each sample and speeds analysis of the resulting data.

Daicel Arbor Biosciences is launching the initial myBaits® Expert Wheat Exome capture panel at the International Plant and Animal Genome Conference in San Diego, CA, USA based on the complete high-confidence exon-annotated genome in hexaploid wheat. The partnership with the IWGSC and its members will continue as Arbor plans to develop new iterations of the exome panel with further understanding of the genome as well as panels specific to disease and drought resistance. Additionally, Daicel Arbor Biosciences is developing a robust bioinformatics pipeline of the exome in order to streamline data analysis and deliver consistency across the consortium.

“We are proud to develop this wheat exome panel, which can be paired with any library preparation, in conjunction with the IWGSC.” explained Jacob Enk, PhD, Senior Scientist at Daicel Arbor Biosciences. “The panel will be added to our targeted sequencing services offering for researchers who require a complete solution from DNA to data analysis.”

The reference sequence for Chinese Spring was made available to the scientific community in January 2017 and an analysis of the reference sequence was published in the journal Science in August 2018. Since the beginning of 2018, more than 250 publications have been published using this resource, yet only a portion of the genome has been manually or functionally annotated with high confidence. Daicel Arbor Biosciences and the IWGSC will continue to collaborate with consortium members as well as outside breeders, plant scientists and wheat growers to further define the genome and develop new resources for studying this valuable crop.
